Protoplast Isolation Device



We designed a microfluidic device for traping and isolating protoplast cells as well as automatedly switching media. The device consists of a bulk channel with a "v-shaped" pillar array for trapping the protoplast cells. Cells were spatially isolated and imaged using fluorescent microscopy to track fusion-protein localization within cells. The device allows cell trapping despite debris heavy solutions (minimizes trapping debris since solution is difficult to purify) and is gentle enough for trapping protoplasts. Reagents could be switched from three inputs, allowing automation and media pertubations.
